A few things to know.

Screen sampling depends on browser support and requires your action. Image sampling reads the browser-decoded image in sRGB; it is not a print-proofing workflow. Images are limited to 24 megapixels and 25 MB.

A little more clarity.

Why is screen sampling unavailable in my browser?

The EyeDropper API is not supported everywhere. Manual controls and local image sampling remain available.

Can I sample transparent pixels?

Choose a sampling backdrop or enable Keep sampled alpha to inspect the original pixel transparency. The preview is browser-decoded sRGB; palette extraction always summarizes colors on its chosen backdrop.
